Glucocorticoid regulation of cell survival in normal and cytokine-treated gastric chief cells
نویسندگان
چکیده
Gastric chief cells secrete pepsinogen and are found at the base of gastric glands. Chief cells constitutively express the anti-apoptotic protein Bcl-xL, which may enable them to live longer than other gastric cells. We reveal that the cytokines released as part of the inflammatory response during Helicobacter pylori infection dose-dependently disrupt chief cell monolayer integrity and viability. We also show that glucocorticoids dose-dependently increase expression of Bcl-xL, and hypothesize that this accounts for the ability of glucocorticoids to protect chief cells from cytokine-induced cell death. In addition, we show that glucocorticoids restrict the ability of cytokines to damage chief cell monolayer integrity. Finally, we propose two mechanisms by which cytokines overwhelm the protective ability of glucocorticoids, resulting in chief cell deletion and gastric atrophy during Helicobacter pylori infection.

Cancer is one of the main causes of mortality. Therapeutic importance in some diseases, especially cancers has led to considering the anti-cancer effects of probiotics more than ever. Probiotics have stimulatory effects on the immunity and suppressor effects on cancer activity. Various studies report that probiotic bacteria have cytotoxicity effects on different types of solid tumors. In this r...
متن کاملEffect of Hesperetin on the level of reactive oxygen species (ROS) in gastric cancer stem cells: Short Communication
Intracellular reactive oxygen species (ROS) play an important role in cancer stem cell (CSC) function. Hesperetin (Hst) is a flavonoid that has been shown to affect cellular ROS level. The goal of this study was to investigate the effect of Hst on the level of ROS in gastric CSCs (GCSCs). MTT assay was used to evaluate cell survival. Cellular ROS level was measured using 2′,7′-dichlorofluoresci...

Background & Aim: Tolerance to the semi-allogenic fetal graft by the maternal immune system is a medical enigma that has stimulated investigations for a half of century. Several hypotheses have been proposed to explain the tolerance of mother to the fetus. The successful pregnancy is proposed and proved by many scientists to be a Th2 dominant phenomenon. This hypothesis is proved in most as...
متن کاملKnockdown of mitofilin inhibits autophagy and facilitates starvation-induced apoptosis in HeLa cells
Objective(s): Mitofilin contributes to the maintenance of mitochondrial structure and functions. This study was undertaken to determine the mechanisms underlying its regulation of apoptosis. Materials and Methods: Mitofilin was knockdowned by specific short hairpin RNA (shRNA) and the stable HeLa cell clone was selected. The autophagy a...

Introduction Insulin-like growth factor I (IGF-I) which is produced in abundance in the normal adult liver, is deeply involved in hepatocyte survival, growth, and differentiation during liver development. IGF-I plays the roles via the receptor (IGF-IR) signaling pathway. IGF-IR unlike IGF-I is expressed strongly in the developing liver, but much more weakly in adults. Objective: We hypothesi...
